BFGF-cDNA transfected iris pigment epithelium cells rescue photoreceptor cell degeneration in RCS rats

Purpose Recently some institutes have been reported to try retinal pigment epithelium (RPE) transplantation for age related macular degeneration (ARMD) They have used RPE obtained from eye bank eye or fetus eye, but m sucn cases, we must always be careful for tissue rejection IT iris pigment epithelium (IPE) couid work as a substitute for RPE, it must be very helpful for promoting transplantation because we can use autograft of ARMD patient themselves We investigated the effect of IPE transplantation on the retinal degeneration in dystrophic RCS rat retina. Method IPE was obtained from adult Long Evans rats We inserted rat bFGF-cDNA into a high expression vector. pCXN2 and iransfected it into cultured rat IPE mRNA was extracted from 3000 cells of pCXN2 bFGF or pCXN2 transfected IPE eacli, and single strand cDNA was synthesized by reverse tianscriptase with random hexamer. then PCR was performed with bFGF specific pnmer, bFGF-cDNA transfected IPE was injected into the subretinal space of post natal 21 day old RCS rat. These rats were sacrificed 30 days after injection Result The expression of bFGF in bFGF-cDNA transfected IPE was much stronger than that in vector transfected tPE Rescue effect of bFGF transfected IPE is very strong m the site of transplantation, whereas the outer nuclear layer of the other side to tne optic disk was very thin Conclusion Factrovich et ai. demonstrated that photoreceptor was rescued by bFGF injection into the eye of RCS rat If bFGF could be delivered constantly to the photoreceptor cells of RCS rat it could help for decreasing their death The tranplantation of bFGF-cDNA transfected IPE was very effective for surviving photoreceptor cell in dystrophic RCS rat.
