N2 - Turn on the switch: Fluorogenic probes for protein labeling based on the photoactive yellow protein (PYP) tag were developed. The fluorescence of the probes is turned off by intramolecular association and switched on by the reversal of this interaction upon reaction with the PYP tag that is fused to the protein of interest (POI, see scheme). The rapid and specific labeling reaction enabled the imaging of cell-surface proteins without washing.
