Abstract
Changes in the interactions between an antigen and antibody upon single amino acid mutation were directly investigated using colloidal probe atomic force microscopy. Interactions studied were those between hen egg-white Lysozyme (Lysozyme) and HyHEL 10 scFv that is an antibody of Lysozyme and a mutant of the Lysozyme, K97M and K97A. K97M has a small association constant with HyHEL 10 scFv, whilst K97A doesn't associate with Lysozyme. We observed the adhesive force between HyHEL 10 scFv / Lysozyme and between HyHEL 10 scFv / K97M, and the former was larger than the latter. No adhesive force between HyHEL 10 scFv / K97A was observed. This is the first direct demonstration of the changes in the interactions upon a single amino acid mutation.
