Quantitative analysis of bile acid 3-sulfates by LC-ESI-oa-TOF mass spectrometer

Abstract

The quantitative analysis of bile acid 3-sulfates by using liquid chromatography - time of flight mass spectrometry (LC-TOFMS) is described. Recently, the TOFMS is used as a combination with a LC. However, most of LC-TOFMS has a Time-to-Digital Converter (TDC) as a data acquisition system. In case of using a TDC, it has limited dynamic range for sample concentration. Most recently, we developed a new type of LC-TOFMS "AccuTQF™", which has an Analog-to-Digital Converter (ADC) as a data acquisition system. By using an ADC, it is possible to accomplish a quantitative analysis with a wide dynamic range. The high-selectivity quantitative determination is expected, because highest sensitivity is obtained with high mass resolution (>6,000) with a TOF instrument.
