Abstract
Transcription factor protein, Spo0A-DB, of Bacillus Subtilis recognize base sequence TGTCGAA called 0Abox. We measured interactions between Spo0A-DB and DNA at various Spo0A-DB densities using colloid probe atomic force microscope. For this interaction measurement, we found the optimum density of Spo0A-DB which corresponded to the closest packing. At excess adsorbed conditions, Spo0A-DB isn't in an appropriate orientation or the DNA binding site of SpoOA-DB is blocked by others, and at smaller densities, influences of the bottom chelate layer is increased.
