The influence of anticoagulant and temperature in measurement of plasma hemoglobin concentration for hemolysis test

Abstract

Hemolysis test has been used to evaluate medical device safety and hemocompatibility. However, the influence of measuring plasma hemoglobin concentration could be very complex, making it difficult to get reproducible hemolysis index. The purpose of this study is to compare the variation of plasma hemoglobin concentration by using 3 kinds of anticoagulants (EDTA-2Na, heparin, sodium citrate), and the influence of temperatures(25°C and 37°C). As a means to measure hemoglobin concentration, Cripps method and Porter method were used. The results showed that no significant decrease in hemoglobin concentration was observed in heparin and sodium citrate. However, the hemoglobin concentration in EDTA-2Na decreased. At 37 °C, the decrease in hemoglobin concentration was also observed. Meanwhile, no significant decrease in hemoglobin concentration was observed at 25°C. The reason for the decrease in hemoglobin concentration is considered to be the formation of methemoglobin.
